Custom Expression Plamids
Working with a client’s scientists and intellectual property
lawyers, Walker Downey and Associates will develop novel
expression plasmids that produce high yields of recombinant
proteins and are free from infringing intellectual property
concerns. We will produce a series of plasmids with different
regulatory elements (promoters, introns, untranslated regions,
polyadenylation regions, etc) that perform well in your cell
type and may be patented, thus protecting this key component of
your production system.
Tailoring Client Supplied Plasmids
Working from a client supplied plasmid, Walker Downey and
Associates will customize the plasmid to function as a discovery
tool. The possibilities are endless, but examples include
tailoring the plasmid to accept mRNA pools creating expression
libraries, or to allow interchangeability of regulatory elements
within a plasmid, useful for optimizing expression of new target
proteins by promoter screening.
Recombinant Services
The
construction of expression plasmids is near the front of the
product pipeline. Many candidate proteins are needed to fill the
pipeline because there are few viable candidates that emerge
from the pipeline’s end. Walker Downey and Associates can
produce the expression plasmids a client needs to feed the
pipeline, freeing the client’s staff to pursue the downstream
identification and characterization of lead candidates.
Troubleshooting
Sometimes expression plasmids just don’t work as well as hoped.

analyses and literature searches to find reasons a client’s
plasmids aren’t performing, and provide detailed suggestions for
getting the plasmid to perform up to expectations.
